About the job Senior Protein Engineer
Key Responsibilities include:
Conceptualize, design, construct, and implement methodologies for novel enzyme and protein discovery
Periodically research literature to focus and supplement research on metabolic and signaling interventions for mammalian & non-mammalian animal cells
Directly support proteomics work by providing expertise on design, post-translational modifications and testing of proteins and protein analogs
Improve kinetic properties of enzymes
Increase stability and biological activity of proteins
Perform techniques such as SDS-PAGE,
Perform assessments for protein engineering method selection including site-directed mutagenesis, DNA shuffling & chemical modification on a case-by-case basis
Perform mechanistic characterization of protein activity and stability
Maintain up-to-date expertise in the field across technologies relevant to the role
Independently advance projects through collaboration with a diverse team of wet-lab scientists, engineers, and computational scientists
Test the performance of the engineered strains using growth, metabolic, and productivity assays including HPLC, ELISA, SDS-PAGE, Western Blotting, etc.
Collaborate cross-functionally to plan and execute demanding cell engineering and screening campaigns that produce strains to meet bioprocess needs and GMP requirements
Interpret and present experimental data to scientific, engineering, and leadership teams
Design and implement screening or selection methods to quickly validate new constructs
Utilize bioinformatics DNA sequence analyses to design and validate successfully engineered strains
Conceptualize, design, construct, and implement platforms for engineering of the proteins e.g, multi-specificity, stability, conformational restraint, effector function, expression and framework optimization
Keep abreast of scientific literature in the field and implement novel modalities and protocols.
Provide expertise in evaluation of new platform technologies.
Proactively search for novel technologies or product opportunities and establish collaborations and research programs to deliver innovative technical solutions.
Design and construct cell strains via optimization of proteins in cell-free environments based on understanding of cell metabolism, review of relevant literature, and collaboration with scientific leadership.
The ideal candidate will possess the following qualities & qualifications:
Doctoral degree (PhD or equivalent) in a related scientific discipline + 4 years of research experience or MS degree in related field + 10 years of research experience
Experience with rational or screen-based engineering to optimize protein activity
Experience with cell-based assays
Track record of moving projects forward
Experience with CRISPR-Cas and/or Antibody engineering, expression, purification, and characterization
Experience with chemical and/or bioconjugation methods and related analytical techniques
Experience in a variety of chromatography techniques including affinity, IEX, and SEC
Experience or a high level of familiarity with NGS-based analytical techniques
Experience or a high level of familiarity with genome editing
Experience or a high level of familiarity with animal metabolism and signaling pathways
